1 助成番号 0906 駿河湾深層水からの海洋微生物の単離とその産生する有用物質の単離 小谷真也, 河岸洋和 静岡大学創造科学技術大学院 概要本研究の目的は 駿河湾の深層水から海洋微生物を単離しカルチャーコレクションを作り そこから新規有用物質を単離することである 駿河湾深層水の利用は 静岡県によって推進されているプロジェクトであり 焼津市沖の駿河湾から定期的に海洋深層水を取水している 深層水は 気候変動や人間活動の影響を受けないため 遺伝的に大きく異なる微生物が生息している可能性が考えられる 現在 水深 397 メートル ( 黒潮系 ) 及び 687 メートル ( 亜寒帯系 ) の 2 層から取水しており 多様な微生物が得られることが予測され この深層水から海洋微生物を単離し発酵産業のために利用することを目指す 駿河湾深層水から 約 50 株の菌株を単離し カルチャーコレクション化した それぞれの菌株を再び寒天培地で培養を行い 抽出用のサンプルを作成した アセトンを用いてそれぞれの菌株を培地ごと抽出した アセトンを濃縮後 ジメチルスルフォキシドに再溶解し 抗菌試験に付した 抗菌試験は 大腸菌 Escherichia coli 黄色ブドウ球菌 Staphylococcus aureus 枯草菌 Bacillus subtilis の 3 種類を用いて行った その結果 駿河湾の深層水から得られた と命名したカビの抽出液が枯草菌 Bacillus subtilis に対し 顕著な抗菌活性を示した そこで 三菱化学疎水性樹脂 CHP20P を用いた逆相クロマトグラフィーで溶媒分画を行ったところ 100% MeOH 画分に抗菌活性が見られた そこで この 100% MeOH 画分から HPLC を用いて 活性物質の単離を試みた HPLC は アセトニトリル / 水の溶媒系で ODS カラムを使用した グラジエントモードで分析を行い 保持時間 分のピークに抗菌活性が見いだされた そこで 大量分取を行い 物質の収量 10.5 mg を得た そこで ESI-MS の測定を行った 陽イオンモードで に陰イオンモードで にイオンピークが観測された また 詳細な構造解析を行うため 核磁気共鳴装置 JEOL ECA600 による解析を行った サンプルは 重クロロホルムに溶解し 測定を行った 1 H-NMR の結果 メチルプロトン 多数のメチレンプロトン オレフィンプロトンが観測された また さらに詳細な構造解析を行うため 二次元 NMR の測定を行った COSY HMBC HMQC 法を用いて測定を行った結果 カルボニルカーボンの存在が明らかになった これらの結果を総合すると 本物質はリノール酸であることが同定された 目的わが国は周囲を海に囲まれ 水圏資源に恵まれており その活用が産業的に重要視されている 特に 太平洋側には数千メートルの深海が存在するが その利用に関してほぼ手つかずである 人類が宇宙にロケットを飛ばす時代になってさえ 深海は未知の世界であり 地球上に残された最後のフロンティアである また 深海生物の中で 好圧細菌と呼ばれる種類の細菌が存在し 水圧 500 Pa 以上でないと生育不能という生物学的に興味深い生理を有 する細菌が発見されている このように陸上とはかけ離れた環境で何億年という時間を過ごしているため 異なる進化を遂げていることが示唆されている 一方で 人類はストレプトマイシンを始めとする微生物由来の抗生物質を発見し その恩恵にあずかってきた 1900 年代初頭から微生物由来の抗生物質の探索研究は世界中で競って行われており 現在までに発見された抗生物質の数は 4,000 を超え 50 以上のものが臨床的に使用されている わが国では昔からみそなどの食品発酵技
2 術が進んでいるため 抗生物質の分野でも競争力を持ち 抗生物質生産は医薬品のなかでも上位を占めている (1994 年日本の医薬品の生産高 5 兆 7,500 億円のうち抗生物質は約 4,000 億円 ) 抗生物質の発見により 人類は細菌感染症を克服し 癌治療においても抗腫瘍抗生物質を用いて治療が行えるようになった これらの抗生物質の約 60% が放線菌と呼ばれる細菌によって産生されている 放線菌は主に土壌中に存在し これまで多種多様な放線菌が発見され 生物資源として用いられてきた しかしながら 世界中の研究者が競って放線菌の単離を行って来たため 土壌から新種の放線菌は発見が困難となり それと共に新奇骨格を持つ二次代謝産物の発見も減少している そこで 現在注目されている生物資源は海洋放線菌である 海洋の底土には放線菌やカビの胞子が付着していることが古くから知られており 最近グアムなど熱帯の海洋底土から Salinispora 属という放線菌の一群が発見された (Jensen PR et al. Environ Microbiol. 2005) さらにこの属の放線菌から抗腫瘍物質 Salinosporamide が単離され 注目を集めている その生産菌 Salinispora tropicana の全ゲノム解析も行われ 遺伝子レベルで陸上の放線菌と大きく異なっていることが報告された (Udwary DW et al. PNAS 2007) 海洋底土には多くの放線菌種が存在し まだまだ未発見の放線菌が存在するといわれている このような海洋細菌に注目が集まっている背景のもと 静岡県の地理的独自性を生かし 地元の生物資源の開発を行うことを着想した 本研究の目的は 駿河湾の深層水か ら海洋微生物を単離しカルチャーコレクションを作り そこから新規有用物質を単離することである 駿河湾深層水の利用は 静岡県によって推進されているプロジェクトであり 焼津市沖の駿河湾から定期的に海洋深層水を取水している ( 図 1) 深層水は 気候変動や人間活動の影響を受けないため 遺伝的に大きく異なる微生物が生息している可能性が考えられる 現在 水深 397 メートル ( 黒潮系 ) 及び 687 メートル ( 亜寒帯系 ) の 2 層から取水しており 多様な微生物が得られることが予測され この深層水から海洋微生物を単離し発酵産業のために利用することを目指す 本研究では水深の深い駿河湾の地理的条件を利用し 新たな生物資源の開発を狙う 本研究において有用な物質が海洋微生物から得られれば 駿河湾深層水の生物資源に注目が集まり 研究のさらなる発展が期待できる 方法焼津市にある駿河湾深層水取水供給施設で平成 20 年 10 月に深層水を採取した 駿河湾深層水取水供給施設の説明平成 13 年 9 月 埋立整備が進められている新焼津漁港 ( 焼津市鰯ケ島 ) に 陸上から海洋深層水を取水するための 駿河湾深層水取水供給施設 が完成した 駿河湾深層水取水供給施設は 海洋深層水の取水と各施設への送水及び一般への給水のための設備 ( 取水ピット, 受水槽, 送水ポンプ, 小口給水所, 大型車両対応給水所 ) を備えている ここでは 駿河湾に取水管を伸ばし 水深 図 1. 静岡の地理と駿河湾深層水
3 397 m( 取水管約 3 km) 687 m( 同約 7 km) の起源の違う 2 種類の海洋深層水及び 試験研究用の表層水 ( 水深 24 m) を取水している 隣接する静岡県水産技術研究所では 駿河湾深層水を用いた魚介類の種苗生産研究や 磯焼け対策のための大型藻類育成の研究等を行っている 駿河湾深層水取水供給施設の西側にあり 平成 16 年度から本格稼動している ( 図 2) 採取した水深 397 m 687 m の深層水を それぞれ海水をベースにした寒天培地上に塗布し 30 で約 7-8 日間 保温し 生育してくる微生物を観察した ( 図 3) 397 m のサンプルで CFU/0.5 ml であり 687 m のサンプルでは 1-2 CFU/1 ml であった すなわち これは 深層水の微生物が非常に少ないことを意味している また 同時に 共同研究者である静岡大学理学部道林准教授から分与を受けたしんかい 6000 で採取したマリアナ海溝深度 6,000 メートルの深海の底土もまたこの方法で存在する微生物の生育を試みた 図 2. 静岡の深層水水産利用施設 実験手法の説明図 1. 環境水を無菌的に寒天培地に撒く 2. スプレッダーで広げる 3. 恒温器で保温する 4. 生育したコロニーを単離する 図 3. 深層水からの菌の単離
4 結果 1. 駿河湾深層水の微生物から抗菌物質の単離約 50 株の菌株を単離し カルチャーコレクション化した それぞれの菌株を再び寒天培地で培養を行い 抽出用のサンプルを作成した アセトンを用いてそれぞれの菌株を培地ごと抽出した アセトンを濃縮後 DMSO に再溶解し 抗菌試験に付した その結果 駿河湾の深層水から得られた と命名したカビの抽出液が活性を示した ( 図 4) そこで 三菱化学疎水性樹脂 CHP20-P を用いた逆相クロマトグラフィーで溶媒分画を行ったところ 100% MeOH 画分に抗菌活性が見られた そこで この 100% MeOH 画分から HPLC を用いて 活性物質の単離を試みた ( 図 5) 100% MeOH 画分の HPLC 分析の結果を下に示す ( 図 6) 非極性部 (20 分以降 ) に顕著なピークが見られた 保持時間 分のピークに抗菌活性が見いだされた そこで 大量分取を行い 物質の収量 10.5 mg を得た そこで ESI -MS の測定を行った ( 図 7) 抗菌物質の 1 H および 13 C NMR の測定を行った 結果を下に示す ( 図 8 及び図 9) さらに詳細な構造解析を行うため 二次元 NMR の測定を行った ( 図 10 および図 11) さらに HMBC スペクトラムによってカルボニルカーボンの帰属が行えた ( 図 12) これらの NMR および ESI-MS スペクトラムの解析の結果 活性物質は下のような構造のリノール酸であると同定された ( 図 13) 図 4. 単離したカビ カビ を寒天培地で培養 アセトンで抽出 アセトン抽出物 20%MeOH 画分 CHP20P 60%MeOH 画分 図 5. 単離したカビ %MeOH 画分 Bucillus subtilis に対する抗菌活性!! 図 6.100% MeOH 画分の HPLC チャート HPLC 分析条件 H 2 O/MeCN の 2 液を混合するグラジエントシステム 0.05% TFA 入り グラジエント条件 5 分間 40% MeCN イソクラティックで固定 5 分後から 25 分間かけて 40% MeCN から 99% MeCN へカラム ODS-A 4.6x250 mm UV 検出波長 215 nm

9 No.0906 Isolation of Marine Microorganisms from Deep Sea of Suruga Bay Shinya Kodani, Hirokazu Kawagishi Graduate School of Science and Technology, Shizuoka University Summary The purpose of the present study is to isolate the marine microorganism from the deep water of Suruga Bay, to make the culture collection, and to isolate a new, useful material. The use of Suruga Bay deep water is a project being promoted by Shizuoka Prefecture, getting water regularly from Suruga bay in the Yaizu City offing. Because the human activity do not influence the deep water, the possibility that a greatly diverse microorganism lives is thought. Various microorganisms should be obtained by getting water from two layers (397 meters and 687 meters in depth). We aim to isolate the marine microorganism from this deep water in order to utilize it for the fermentation industry. The 50 strains of microorganisms were isolated from Suruga Bay deep water. Each strain was cultured with the nutrient agar again, and the sample for the extraction was made. Each strain of each medium was extracted by using the acetone. It dissolved to dimethyl-sulfoxide, after the acetone was evaporated, and it was subjected to the antibacterial examination. The anti-bacterium examination against three kinds of bacteria including Escherichia coli, Staphylococcus aureus, and Bacillus subtilis was accomplished. As a result, the extracted material of the fungi named showed remarkable anti-bacterium against Bacillus subtilis. The separation was performed by the reversed phase open chromatography with Mitsubishi Chemical hydrophobic resin CHP20P. The anti-bacterial activity was seen in 100% MeOH fraction. Then, the isolation of the active compound was accomplished by using HPLC from the 100% MeOH fraction. The anti-bacterial activity was observed in the peak at the retention time of minutes. To elucidate the chemical structure, ESI-MS was measured. The ion peak was observed in m/z in the positive ion mode and m/z in the negative ion mode. In order to analyze details of the structure, the compound was subjected to the analysis using nuclear magnetic resonance device JEOL ECA600. The sample dissolved to chloroform-d for NMR experments. As a result of 1H-NMR, a methyl proton, many methylene protons, and the olefine protons were observed. Moreover, to analyze details or more and the structure, two dimensional NMR were measured. The existence of the carbonyl carbon was clarified as a result of COSY, HMBC, and the HMQC methods. The compound was identified as a linoleic acid
