CHEMOTHERAPY APR.1994 Intraocular pharmacokinetics norfloxacin (NFLX) in pigmented animals Yukihiko Tokushima Mitsui University Hideo Ohkubo Masakatsu Komuro Central Research Laboratories, Kyorin Pharmacentical Co., Ltd., 2399-1, Mitarai, Nogi-machi, Shimotsuga-gun, Tochigi Japan The intraocular pharmacokinetics norfloxacin (NFLX) after 2 weeks consecutive oral administration or instillation [14C]-NFLX in beagle dogs pigmented rabbits were studied for 6 months. The following results were obtained. 1) The concentration radioactivity in choroid pigment epilium (choroidpigment epilium) dogs was 433 pg eq./g at 24 hours, 276 Đg eq./g at 1 month, 89.6 Đg eq./g at 6 months after last 30 mg/kg administration [14C]-NFLX. 2) The concentration in choroid-pigment epilium rabbit was about 90.3 Đg eq./g at 24 hours after last 50 mg/kg administration [14C]-NFLX 3) Concentrations in non-pigmented tissues were lower than serum level. 4) In case instillation with 50 pl 0.3% [14C]-NFLX at 5 times/day for 2 weeks, concentration in iris-ciliary body choroid-pigment epilium dogs were 6.74 2.03 Đg eq./g, respectively, at 24 hours after last application, se radioactivities decreased slowly reafter. 5) Microautoradiography showed localization radioactivity only in uvea, without showing diffusion into retina. These results indicate that NFLX has an affinity for melanin, results in a high accumulation retarded residue in melanotic ocular tissues. Fluoroquinolones are known to have a strong affinity for melanin. Lomefloxacin levloxacin are reported to accumulate in uvea beagle dogs at level 1,200 Đg/g. In monkey, fluoroquinolone accumulation 138-300 Đg/g is reported. Based on se findings, long-term use fluoroquinolones should be considered in order to avoid melanin disorder.
