JONES et al. : Prophylaxis and therapy for Pseudomonas aeruginosa infection with carbenicillin and with gentamicin. Brit. Med. J. 3 : 79-82, 1967 OPHTHALMIC USE OF CARBENICILLIN MASAKICHI MIKUNI, MASAO OHISHI, SHIGEO SUDA, MASAO IMAI & TAKAKO TAKAHASHI Department of Ophthalmology, Niigata University School of Medicine (Director : Prof. M. MIKUNI) Bacterial and clinical experiments for ophthalmic use of carbenicillin (CB-PC) were performed and the results summarized as follows. 1) Minimum growth inhibitory concentration of CB-PC was mcg/ml for K-W bacillus, mcg/ ml for M-A bacillus, '25 mcg/ml for Pneumococcus, mcg/m1 for C. diphtheriae, mcg/ ml for Gonococcus, mcg/ml for Streptococcus, mcg/m1 for Staphylococcus and racg/m1 for Ps. aeruginosa,

10 1306 CHEMOTHERAPY AUG ) The distribution of the sensitivity for 100 strains of Staph. aureus was in the range of mcg/ml, and the peak existed in 5 mcg/ml. 3) Fifteen strains of Ps. aeruginosa were sensitive in 25,--->100 mcg/m1 : 25 mcg/ml for I strain, 50 mcg/ml for 5 strains, 100 mcg/ml for 7 strains. and >100 mcg/m1 for 2 strains. 4) The blood level by intramuscular injection of a single dose 1 g reached the highest after 1 hour. and decreased gradually until 6 hours. 5) After the intramuscular injection of CB-PC in a dose of 50 mg/kg, the concentration of ocular tissue was found in the outer and inner parts of the rabbit eye. After instillation of I % CB-PC eyedrop, the concentration was only obtained in the outer part of the eye. After 5 mg/0. 5 ml subconjunctival injection was recognized not only in the tissue of the outer segment, but also in the inner parts of the eye. 6) No disturbance of the eye was observed by the application of CB-PC eye-drop, ointment and subconjunctival injection. 7) CB-PC eye-drop was stable and the antibacterial power was not decreased for 10 days. 8) Intramuscular injection of 1 g CB-PC once or twice daily revealled excellent effects on 11 cases of hordeolum, 2 cases of acute chalazion, each one case of lid abscess, acute-, and chronic dacryocystitis, 3 cases of ulcer of the cornea, and a case of orbital abscess. 9) Side effects : Some of patients experienced pains on injection, but any otner severe side effect, such as allergic reaction, were not noticed.
