JAPANESE JOURNAL OF INFLAMMATION ORIGINAL ARTICLE Effects of acteoside (TJC-160) on the expression of tissue inhibitor of matrix metalloproteinase(timp) in glomeruli of crescentic-type anti-gbm nephritic rats In order to clarify the antinephritic mechanisms of TJC-160, we investigated the effect on the increased expression of TIMP in glomeruli of crescentic-type anti- GBM nephritic rats. We administered TJC-160 (30mg/kg/day, p.o.) to the rats from the 20th day to the 35th day after anti-gbm nephritis induction. TJC-160 suppressed proteinuria and total/fibrous crescent formation in glomeruli. TJC-160 suppressed the increased expression of TIMP in crescent, compared to vehicle injected control nephritic rats. In addition, TJC-160 inhibited the decreased activities of metalloproteinase (MMP)-1 and 2,9 in glomeruli. Despite the inhibition of the decreased MMP activities, TJC-160 did not enhance the MMP activities in normal glomeruli in the in vitro study. TJC-160 had no effect on the activity of MMP in trypsin treated glomeruli of nephritic rats at the 25th and the 35th days. These data show that the effect of TJC-160 to inhibit the process of fibrous crescent formation in anti-gbm nephritic rats may be partly due to the ability of TJC-160 to prevent TIMP expression in nephritic glomeruli. (2)半 月体 形 成 に対 す るTJC-160の 腎 炎 対 照 群 で は25日 目 に 約50%の 月体 形 成 が 観 察 され,35日 糸球 体 に半 目に は約70%ま で増 加 した.TJC-160投 与 に よ り25日 目 な らび に35日 目 に ほ ぼ50%の 抑 制 率 で 半 月体 形 成 が 抑 制 さ れ た.さ ら に線 維 化 半 月 体 は25日 目 に は50%の 目 に20%,35日 糸 球 体 に 認 め ら れ た が,TJC-160 投 与 群 で は70 80%の 抑 制 率 で有 意 な抑 制 が 見 られ た. 線 維 化 半 月 体 形 成 率 に対 して もTJC-160投 群 で は35日 (3)TIMP発 現 に対 す るTJC-160の 図3に35日 た.腎 与 目 に有 意 な 低 下 を示 した(図2). 目の 糸 球 体TIMPの 炎 対 照 群(図3A)の 染 色 像 を示 し ほ とん どの 糸 球 体 で は 半 月 体 部 位 に陽 性 に染 色 され て お り,正 常 群 で は 痕 跡 が陰 性 で あ っ た(写 真 未 発 表).TJC-160投 群(図3B)で の,腎 与 は 半 月 体 部 位 に染 色 さ れ て い る も の 炎 対 照 群 に比 較 して 軽 度 で あ っ た.以 上 の 所 見 を ま と め て 図4に 示 した.25日 日 目 に は 糸 球 体 の 半 月 体TIMP発 し く増 加 した.TJC-160投 目 な い し35 現 はいち じ る 与 群 で は25日 制 傾 向 で あ っ たが,35日 目 に抑 目 に は有 意 な抑 制 が 認 め られ た. (4)糸 球 体MMP活 性 に対 す るTJC-160の MMP-1活 性 は25な い し35日 目 で正 常 値 に く らべ て 腎 炎群 で い ち じ る し く低 下 した.TJC-160 投 与 群 に お い て は25日 目 に は 回復 傾 向 を示 し,35 日 目 に は ほぼ 正 常 レベ ル まで 改 善 が 認 め られ た. MMP-2,9活 与 群 で25な いし 35日 目 にお い て有 意 な改 善 が 認 め られ た(図5). vitroで の 糸 球 体MMP活 TJC-160の 性 に対 す る in vitroで はMMP-1活 の100μM処 ト リプ シ ン処 置 に よ る総MMP活 た が,25日 目 な い し35日 目 に お い て 正 常 群 と対 照 群 との 活 性 の差 は認 め られ な か っ た.TJC-160 投 与 群 で35日 性 に対 してTJC-160 性 を測 定 し 目 に活 性 の 亢 進 傾 向 を 示 した の み で あ っ た(図7). 置 に お い て の み有 意 な活 性 亢 進 が 認 め られ た が,MMP-2,9活 性 に は影 響 が み られ な か った(図6).
