Species identification and genetic diversity of Meloidogyne javanica on pistachio in Kerman province, Iran Meloidogyne javanica is the most widespread species of root-knot nematode on pistachio in Kerman province, Iran. In order to identify the species and investigate the species genetic diversity, during 2004, 93 root-knot nematode populations and related soils collected. M. javanica populations identified based on morphological features, differential hosts and species-specific primer pairs. The results showed 88% of whole collected populations were M. javanica. RFLP profiles from AluI, Tru9I, BamHI, PstI and TaqI digests of 670 and 1600 bp fragments confirmed that populations are identical. In RAPD analysis, that covers the expanded area of the genome, 10 out of 30 primers showed polymorphism among M. javanica populations. The most polymorphic profile was obtained using OPA-10 with 14 bands. Cluster analysis of RAPD profiles was performed using NTSYSpc V based on UPGMA method and dice coefficient. Populations of M. javanica classified in five groups based on 95% similarity level. The result revealed percent similarity among different populations of M. javanica. M. Olia (Dept. Plant Prot., Agr. Nematological Research in Iran There are different climatic conditions for the cultivation of crop plants in Iran and the nematode fauna varies considerably in different parts of the country. More than 200 plant parasitic and free living nematodes from different orders have been reported from Iran of which root-knot nematode species mainly Meloidogyne javanica and M. incognita on wide range of crops like vegetables, pomegranate and pistachio, Heterodera schachtii on sugar beet and Tylenchulus semipenetrans on citrus trees are of paramount economic importance. Attempts have been made to reduce the nematode population under the economic threshold level using cultural, biological and chemical practices such as planting trap crops like Raphanus raphanistrum, application of nematode antagonistic fungi like Paecilomyces spp. and nematicide application such as Temik, respectively. A Teratorhabditis species isolated from red palm weevil, Rhynchophorus ferrugineus The dauer juveniles of a species of rhabditid nematode were isolated from underneath the elytra of the red palm weevil, Rhynchophorus ferrugineus, from a dead Canary Island date palm, Phoenix canariensis in Kagoshima, Japan. The dauer juveniles were cultured on a NGM agar plate, and the adult nematodes were observed with light microscopy. The morphological features of the adults fit the definition of the genus Teratorhabditis, and also fit a species reported from India as a Pelodera species, although there were several discrepancies. The full length of the ribosomal small subunit (SSU) and D2/D3 expansion segment of the ribosomal large subunit (LSU) were sequenced for molecular profiles of the Teratorhabditis sp. Based upon the morphology and molecular profile, Teratorhabditis sp. is considered closest (sister) to T. synpapillata, although its biological traits were closest to those of T. palmarum. K. L. Study on control of the Japanese pine sawyer, Monochamus alternatus and the red palm weevil, Rhynchophorus ferrugineus by entomopathogenic nematodes In this study, we have screened some species of entomopathogenic nematodes for controlling Japanese pine sawyer, Monochamus alternatus and the red palm weevil, Rhynchophorus ferrugineus such as Steinernema sangi TX1 and Heterorhabditis indica MP11 (isolated from Vietnam), S. feltiae and S. kushidai (isolated from Japan), and S. glaseri and S. carpocapsae (commercial species). The filter paper test in laboratory showed that S. carpocapsae and S. glaseri had good virulence and penetration rates to stage 1-4 of Monochamus alternatus and immature stage of Rhynchophorus ferrugineus but they did not infect to mature stage of both hosts. The migration of S. carpocapsae and S. glaseri were compared in the pine sawdust column with or without M. alternatus at the bottom. The migration of S. glaseri in the column with an insect host was significantly higher than the column without an insect host, and could penetrate to host after migrated 16 cm at 25 C in 24 h. In contrary, the majority of S. carpocapsae remained in the segment of application. Y. (Nematoda: Parasitaphelenchidae) isolated from dead pine trees killed by PWNs In the studies of seasonal changes in nematode fauna in the pine trees killed by PWNs, an undescribed species of Bursaphelenchus was found with other 15 species of nematodes in dying Japanese black pine trees (Pinus thunbergii). The nematode is characterized by an elongate-oval of median bulb, longer than width and one body diameter length, rectum and anus indistinct, intestine terminating in blind sac, male tail strongly ventrally curve, slightly digitate tip, spicule arcuate, condylus rounded, slightly elongated, lamina smoothly curved, rostrum rounded, cucullus present, flattend and distinct of bursa. The nematode is closely related to B. hunanensis, B. minutus, B. sexdentati, B. poligraphi, and B. piniperdae in the shape of spicule but shares some characters with Ektaphelenchidae species, i.e. head well set-of and lips not equal size, posterior position of valve plate in the median bulb, intestine terminating in blind sac and indistinct of rectum and anus. 115
