1 交配後の笑気ガス処理による四倍体シンテッポウユリの作出 佐藤孝夫 抄録 観賞用植物における倍数体では 日持ち性の向上 花色の濃色化 花型の巨大化などの有望形質が報告されている 花きでは さらに新たな形質が求められており 倍数性育種の利用が今後ますます進んでいくと思われる ユリの倍数性育種には これまでにコルヒチン処理が主に用いられてきた しかしながら コルヒチンは変異原作用をもつため キメラ個体の出現などの問題点がある 本試験では コルヒチン処理に代わる染色体倍加法として笑気ガス処理による四倍体の作出方法について検討した 供試品種はシンテッポウユリ品種 雷山 2 号 と 北沢早生 の交配系統で これらの交配後 日目にそれぞれ 6 気圧で 72 時間の処理を行ったところ 交配後 13 日目の処理で四倍体が 95% と最も高頻度で作出された 四倍体の作出条件の品種間差異を確認するために 子房親として 雷山 2 号 優雅中早生 オーガスタ の 3 品種 花粉親として 北沢早生 雷山 2 号 ホワイトランサー の 3 品種を子房親として 任意に 8 通りの組み合わせで交配を行い 交配 13 日後から 6 気圧 72 時間の笑気ガス処理を行ったところ 73~100% の四倍体作出率であったことから シンテッポウユリにおいて交配後の笑気ガス処理は四倍体を効率的に作出できることを明らかにした キーワード : 笑気ガス 四倍体 交配 シンテッポウユリ 1 緒言ユリ (Lilium spp. 2n=24) は花型の多様性やエレガントな香りなどにより世界中で好まれている球根性花きで 主に切り花や花壇用として栽培されている 国内においては切り花として需要が多く ほぼ全国的に栽培されており 平成 25 年度の作付面積はキク類に次いで第 2 位の重要な品目である ( 農水省 2014) ユリの自生地は主に北米大陸 ユーラシア大陸などの北半球に限られて分布し 日本には固有種 9 種 共通種 6 種の合計 15 種が自生している ( 清水 1987) 日本固有種であるヤマユリは花型が優雅で芳香性があるなど 観賞用として秀でており 国内では品種改良の必要性が乏しかったため 他の花きに比べて育種の歴史は短い しかしながら 海外では日本固有種の人気は高く 特にヤマユリは カサブランカ などのオリエンタルハイブリッド系ユリの育種親として利用されている ユリの育種では交雑親和性のあるユリ同士の種間交雑が盛んに行われており 花型 花色 花の大きさなどの様々な形質を表す品種が育成されている さらに多様な形質を求めて 花柱切断技術 ( 浅野 明道 1977a, b) と胚培養技術 (Asano 1980a, b) が開発され それらの組み合わせによる遠縁交雑で雑種ができるようになった さらには染色体倍加法による三倍体や四倍体品種が育成されている 観賞用植物における倍数体の有望形質として 日持ち性の向上 (Ketsa ら 2001) 花色の濃色化(Takamura and Miyajima 1996) 花型の巨大化 (Arisumi 1964) などが報告されており 今後 花きの新たな形質の作出には 倍数性育種の利用が有効と思われる ユリの倍数性育種には コルヒチン処理が主に用いられてきた (Emsweller 1949) しかしながら コルヒチンは変異原作用をもつため キメラ個体が出現しやすく さらに球根性であるユリは 実生から開花までに数年を要し 稔性回復花粉を得るまでにさらに年数がかかるという問題点がある コルヒチン処理に代わる染色体倍加法として細胞の減数分裂期に笑気ガス処理を行う方法が見いだされ (Östergren 1954) これまでにチューリップ(Okazaki ら 2005) やユリ (Akutsu ら 2007) で 花粉の減数分裂期に笑気ガスを処理して倍加花粉を作出した報告がある 笑気ガスは気体のため 加圧して処理すると植物細胞内に容易に入り込み 処理後はただちに細胞外 本研究は 農林水産省高度化事業 新染色体倍加法を用いた種子繁殖および早期開花性ユリの開発 (2007 年 ~2009 年 ) により行い 本報告の一部は 国際園芸学会第 23 回 Eurcarpia シンポジウムにおいて発表した 2015 年 3 月 1 日受付 2015 年 3 月 31 日受理
2 へ放出されるため 処理が容易である しかしながら減数分裂は急速に進行するため処理適期を逸することがあり 倍加花粉を作出する頻度が不安定である さらに倍加花粉を選別する必要があること 倍加花粉を用いた交配で倍加個体を得るには 数年を要することが問題点である しかし 受精卵の体細胞分裂期に笑気ガス処理を行うと 染色体が倍加した種子を短期間で効率的に得られる可能性がある そこで 本県の花き振興作物であるシンテッポウユリを用いて人工交配し 笑気ガスの受精卵処理による四倍体の作出を図った 2 材料と方法試験 1 笑気ガスの処理適期の検討供試品種として子房親に 雷山 2 号 花粉親に 北沢早生 を用いた 播種は 2009 年 2 月 6 日に 200 穴セルトレイに 1 粒まきで行い 無加温ガラス室内のベンチに 20 に設定した電熱線を付設した育苗床にセルトレイを並べて 保温として 0.07mm 厚のビニールフィルムでトンネル被覆した 同年 5 月上旬に本葉 3 枚に展開した実生を 10.5cm 径黒ポリポットに逐次移植し 人工気象室 (Koitotoron, Koito Ind. Co., Ltd., Tokyo, Japan) で 20 一定で 12 時間日長に設定して生育環境を管理した 供試した子房親 花粉親のなかで開花日が同じ個体同士を任意に交配した 交配では 開花前日に子房親を除雄して袋かけを行い 開花 2~3 日後に交配した 交配して および 15 日後にそれぞれ内径 20cm 高さ 100cm の円筒形の耐圧容器 ( 第 1 図 ) に植物体を黒ポリポットのまま 2 個 たさく果が成熟し 先端が割れたころに採種し 5 に設定した冷蔵庫に保管した 染色体数を確認するため 2010 年 2 月 5 日に 200 穴セルトレイに播種した 播種後は 上記と同様な育苗床にトンネル被覆して管理した 発芽後 セルトレイで本葉 2~3 葉期に達した生葉を用いて 葉を長さ 10~20mm の大きさで切り取り 9cm プラスティックシャーレ上に置き 核を抽出するために A 液 (CyStain UV 植物 DNA 分析試薬 A 液 Partec 製 ) を 0.5ml 加え 剃刀を用いて葉脈に反って細かく刻み 10 分間静置した その後 Cell Trics フィルターに通したプラスチック試験管に滴下して 抽出液を得た さらに抽出液の 4 倍量の B 液 (CyStain UV 植物 DNA 分析試薬 B 液 Partec 製 ) を用いて Cell Trics フィルターに通して滴下して抽出し 氷を入れた 25 20cm 発砲スチロール内で 10 分間以上静置した 流速を 1.0μl/s に設定したフローサイトメータ (Ploidy Analyzer (PA) Partec 製 ) を用いて 交配後日数別にそれぞれ 40 個体の相対的 DNA 量を測定し 倍数性を検定した 内部標準にはシンテッポウユリ 雷山 2 号 を用いた 試験 2 笑気ガス処理による倍数体作出に関する品種間差異供試品種として子房親に 雷山 2 号 優雅中早生 オーガスタ の3 品種を用い 花粉親に 北沢早生 雷山 2 号 オーガスタ ホワイトランサー 優雅中早生 はつき の 6 品種を用いてそれらの品種の中で任意で 8 通りの交配組み合わせを行い 試験 1 と同様に播種 栽培管理を行った 笑気ガス処理は交配 13 日後に 6 気圧 72 時間の条件で行った 処理後は耐圧容器から植物体を取り出し 無加温ガラス温室で管理した 同年 10 月以降 形成されたさく果が成熟し 先端が割れたころに採種し 冷蔵庫に保管した 2010 年 2 月 5 日に 200 穴セルトレイに播種した 播種後は 上記と同様な育苗床にトンネル被覆して管理した セルトレイで本葉 2~3 葉期に達した生葉を用いて 交配後日数別にそれぞれ 20~25 個体を試験 1 と同様にフローサイトメトリー分析により 相対的 DNA 量を測定し 倍数性を検定した 第 1 図笑気ガス処理の耐圧容器 A: 耐圧容器 B: 笑気ガスボンベ Bar=10cm 体入れ 6 気圧の条件下で 72 時間笑気ガス処理を行った 処理後は耐圧容器から植物体を取り出し 無加温ガラス温室で栽培管理した 同年 10 月以降 形成され 3 結果試験 1 笑気ガスの処理適期の検討交配後日数別に 2 個体ずつ笑気ガス処理して獲得した種子を用いて それぞれ 20 実生個体で合計 1 区あたり 40 実生個体のフローサイトメトリー分析を行った 内部標準に用いた 雷山 2 号 の DNA 量の約 2 倍量が検出された個体を四倍体と検定した ( 第 2 図 ) 交配 5 日後処理区ではすべての個体が二倍体であった 交配 7 日後 ~13 日後までの処理区では交配後日数が経過するほど 四倍体の作出数は増加した すなわち交配 7 日後処理区は 40 個体中 8 個体 交配 9 日後処理区
3 は同じく 22 個体 交配 11 日後処理区は 24 個体 交配 13 日後 ( 第 3 図 ) 処理区は 38 個体が作出され 特に交配 13 日後は 95% の高頻度で四倍体が作出された しかしながら 交配 15 日後は 34 個体に減少した ( 第 1 表 ) 細胞数 細胞数 二倍体 四倍体 蛍光強度 第 2 図フロローサイトメトリー分析によるシンテ ッポウユリ四倍体個体の相対 DNA 量 上段 : 標準品種 雷山 2 号 ( 二倍体 ) 下段 : 笑気ガス処理して得られた実生個体 ( 四倍体 ) 山 2 号 ホワイトランサー では 180 粒播種し 任意の 20 個体を検定したところ四倍体は 95% の作出率であった 雷山 2 号 優雅中早生 では 80 粒播種し 任意の 20 個体を検定したところ四倍体は 100% の作出率であった 雷山 2 号 はつき では 110 粒播種し 任意の 20 個体を検定したところ四倍体は 100% の作出率であった 優雅中早生 オーガスタ では 45 粒播種し 任意の 20 個体を検定したところ四倍体の作出率は 100% であった 優雅中早生 ホワイトランサー では播種数 40 粒 検定数は任意の 20 個体で四倍体は 95% であった 優雅中早生 雷山 2 号 では播種数 200 粒 検定数は任意の 25 個体で四倍体は 80% であった オーガスタ 雷山 2 号 では播種数 200 粒 検定数は任意の 30 個体で四倍体は 73.3% であった このようにシンテッポウユリの品種同士の交配による受精卵処理では 品種による差異は認められなかった 4 考察 第 3 図交配 13 日後の笑気ガス処理適期の植物体の姿 Bar = 5cm 試験 2 笑気ガス処理による倍数体作出に関する品種間差異 8 通りの交配組み合わせから種子を得て フローサイトメトリー分析を行った 交配 13 日後の笑気ガス処理により 8 組み合わせの倍数体はそれぞれ 73.3~ 100% と高頻度の作出率であった ( 第 2 表 ) それぞれの内訳は 雷山 2 号 オーガスタ の組み合わせから 100 粒播種し 任意の 20 個体を検定したところ四倍体は 19 個体確認され 作出率は 95% であった 雷 花きは花束として贈答用に用いられたり 冠婚葬祭の飾り花として観賞用に供されたりして 人の心に癒やしを与え 人間の感性を豊かにできるものである しかしながら 花色は流行に左右され 草姿は変化がないと飽きられるため 華麗な色彩や豪華な花型 変化の富む草姿など これまでにない変化に富む形質が求められる 野生種にない新しい形質は 交雑による有用遺伝子の集積やゲノムの倍数体化による集積 自然突然変異など 遺伝的変異によるところが大きく 例えばユリの自生種のオニユリにおいては 二倍体と三倍体 ( 竹中 永松 1930) からなる倍数体複合種のような種もある 染色体の人為的倍加方法については 化学薬剤処理として ユリにおいてはコルヒチン (Asano 1982) やオリザリン (Van Tuyl 1989) が用いられている 一般的には適正な倍率の溶液に侵漬するだけの簡単な処理で染色体を倍加できるコルヒチン処理が行われているが コルヒチンはイネサフランから得られるアルカロイドで毒性があり 突然変異を誘発するため コルヒチンに代わる薬剤としてりん片への
4 オリザリン処理が行われている しかし コルヒチンやオリザリン処理による倍数化個体の作出率は 10~ 20% 程度と低い (Van tuyl 1989) 化学薬品に代わる倍数体の誘発剤として 笑気ガスによる手法が Östergren(1954) により開発された 笑気ガスによる処理はガスを充填する容器が必要になるが 処理後の笑気ガスは急速に拡散され 突然変異の誘発はない 笑気ガス処理による染色体の倍加については 受粉後に笑気ガス処理を行い 同質倍数体として染色体を倍加する手法がクレピス ( キク科 フタマタタンポポ属 Crepis capillaris ;Östergren 1954) で初めて用いられている その試験では 交配 7~11 時間後に 10 気圧で 4 時間笑気ガス処理すると 倍数化個体を含めた異数体の出現が見られて 倍数化個体は 19% の頻度で作出されている 木原ら (1960) は コムギを用いて交配 24 時間後に 6 気圧で 15 時間処理すると倍数体を含む異数体が 98% 出現したと報告されている ほかにアカクローバ ( 松浦ら 1974) では交配 24 時間後に 7.5 気圧条件で笑気ガスを 24 時間処理するのが最も効果的な方法であることが報告されているが 倍数体の作出率は 80% であった さらに 7.5 気圧で 36 時間処理をすると稔実率が低下し また 異常個体の発生率が極めて高くなるので 36 時間などの長時間処理は避けなければならないと指摘されている しかし 本試験の結果では 交配 13 日後の 6 気圧 72 時間の笑気ガス処理で 95% の効率で四倍体が作出され 稔実率の低下や 異常個体の発生は見られなかった 本試験では 交配 5 日後には四倍体個体は認められず その後交配 13 日後までは四倍体個体数は増加し 交配 15 日後の処理開始では 減少するため 交配 13 日後の笑気ガス処理開始が適期と考えられる 交配後 2 週間経過した後の処理開始では 当初 処理時期としては遅いと思われたが シンテッポウユリの柱頭や胚珠の長さが他の花きに比べて長いため 花粉管が胚珠に到達するまでに時間を要すること 胚珠に到達してから胚のうに侵入して重複受精するまでにさらに時間を要することが考えられる さらに他の交配組み合わせで 交配 13 日後の笑気ガス処理を 8 通りの組み合わせで行い それぞれ 73~100% の頻度で四倍体が作出されたので 交配 13 日後の笑気ガス処理がシンテッポウユリでは最も効果的であることを明らかにした ユリは オリエンタルハイブリッドやアジアティクハイブリッドなど多くの品種群で構成されていて 本試験で用いたシンテッポウユリと同様な同質倍数体の作出の可能性があるが 花器の大きさもそれぞれ違うので 品種群別に処理時期を決定する試験を行う必要がある 新たな形質の発現には 同質倍数体よりも異質倍数体のほうが様々な形質が発現しやすいため 笑気ガス を利用した倍数化個体の作出に関しては これまでにチューリップ (Okazaki ら 2005) やユリ (Akutsu ら 2001) において 非還元配偶子の花粉や胚のうを倍加して巨大化した非還元配偶子を作出し これらを交雑して三倍体や四倍体を作出しているが 倍加花粉の出現率は低く 倍加花粉を選別する技術が必要となる とくに倍加した非還元配偶子を 通常の非還元配偶子と区別して巨大花粉を取り出す技術は未確立である 本試験では 笑気ガスを受精卵に 1 回処理することにより 同質四倍体の種子を容易に獲得できるため 育種年限の短縮には効果があり 今後の倍数性育種の手法として大いに役立つと考えられる 5 謝辞本試験の遂行にあたり 有益なアドバイスをいただくとともに 倍数体の検定ではフローサイトメータを利用させていただいた千葉大学大学院園芸学研究科花卉園芸学研究室三吉一光教授 ( 前秋田県立大学生物資源科学部 ) および交配や栽培管理に多大なるご尽力をいただいた農業試験場野菜 花き部非常勤職員上林影子さんには感謝の意を申し上げる 6 引用文献 Akutsu. 6 Induction of Tetraploid Lilium formolongi hort. by treatment of nitrous oxide gas after pollination Takao SATO Abstract Lilium are one of the most favorite and important ornamental flowers in many countries. Tetraploid varieties of ornamental flowers have superior traits such as large flower and higher vase life. Tetraploids of Lilium formolongi hort. could be obtained by soaking the scales in aqueous solution of colchicine. Even if the pollen of tetraploids obtained via colchicine was used to crossing, it takes 2-3 years from sowing to flowering. Furthermore, colchicine is harmful to bulbous plants. Two cultivars of Lilium formolongi hort. namely Raizan No.2 and Kitazawa - Wase were used for the present study. The ovaries of Raizan No.2, which had been pollinated with Kitazawa - Wase pollen grains were treated with nitrous gas for 72h at 6 atm at room temperature in a pressure-tolerant steel cylinder (20cm in inner diameter, 100cm in length). The plants were treated nitrous gas at 5, 7, 9, 11, 13, 15 days after pollination. Flow cytometric analysis revealed that tetraploids were obtained in frequencies at 95% by the treatment for 13 days after pollination. Furthermore, five cultivars of Lilium formolongi hort., namely Raizan No.2 and Kitazawa - Wase, Yuga - Nakawase, Ogasuta and White Ranser were used. These cultivars were pollinated in random, eight cross combinations were gained. The plants were treated by nitrous gas for 13days after pollination, respectively. Flow cytometric analysis revealed that tetraploids were obtained in frequencies at %, respectively. Nitrous gas treatment is useful method for the production of tetraploid lily cultivars.
