Oral Biol., 39: , Effect of reserpine on pilocarpine-induced salivary secretion in mice Hiroshi Saito Department of Pharmacology, School of Dentistry, Iwate Medical University (Chief: Prof. Tadanobu Itoh) Chuodori, Morioka 020, Japan Key words: reserpine/pilocarpine-induced salivation/mouse Abstract: Effects of reserpine on the response to salivation induced by a parasympathomimetic, pilocarpine, in mice were examined. 1. The amount of saliva and the duration of salivation were significantly increased in a dose-dependent the response to pilocarpine-induced salivation 12 hours after the administration of reserpine (1mg/kg) showed a significant decrease compared to the control, whereas the salivary response 7 days after the administration of reserpine recovered to the same level as the control. 2. In response to pilocarpine-induced salivation, the increased salivary response 30 min after administration of reserpine (1mg/kg) was inhibited with a parasympathomimetic blocking agent, atropine, but not 3. The acetylcholine levels in the submandibular and sublingual glands of mice 30 min or 12 hours after the administration of reserpine did not differ from the control levels of the mice 12 hours after reserpine administration showed a significant increase 4. In the reserpinized mice, food consumption, water intake, whereas the choline levels in the glands., and body weight gain showed a reduction in the 1st and 2nd days after reserpine administration, but were recovered 7 days after administration. These findings suggest that in mice receiving a single administration of reserpine, the pilocarpine-induced salivary response displays a time-dependent biphasic magnitude: a dose-dependent increase 30 min afte r the administration of reserpine, and a decrease 12 hours after the administration of reserpine. In addition, this increase in the pilocarpine-induced salivary response suggests that the reactivity of the parasym pathetic nervous system accelerated, because its inhibition was dependent on the dosage of parasym pathomimetic
