1 安田女子大学紀要 一知* The Antioxidant Activity of Camu-camu Drink Mutsuko Shirai, Kazunori T akamura* Abstract This study investigated the antioxidant activity of Camu-camu drink by 1,1-diphenyl-2picrylhydrazyl (DPPH) radical-scavenging activity, analyzing vitamin C and total polyphenol contents. The following results were obtained: 1) The vitamin C content of 60% Camu-camu drink (667mg/100ml) was higher than that of 80% Acerola drink (400mg/100ml). 2) The total polyphenol content of 60% Camu-camu drink (108mg/100ml) was lower than that of 80% Acerola drink (161mg/100ml), and higher than that of green tea (67mg/100ml). 3) The DPPH radical-scavenging activities were high in the order of 60% Camu-camu drink > 80% Acerola drink > green tea > 10% Camu-camu drink > 0.1% vitamin C solution. In particular, 60% Camu-camu drink (10,353µmol DPPH trapped/100ml) had DPPH radicalscavenging activity that was about twice higher than 80% Acerola drink (5,694µmol DPPH trapped/100ml). 4) The result of this research clarified that 60% camu-camu drink contained high vitamin C and polyphenol, and possessed the antioxidant activity. 2 326 一知 推奨しているが 現地で一次加工したもの以外は輸出禁止になっている 最近注目され始めた果 物であるカムカム果汁の抽出４ や果実中の成分を定量した研究報告はあるが５ ６ カムカム飲 料の抗酸化活性を測定した研究報告は少ない 本研究では を検討す るために ビタミンＣ含量 総ポリフェノール含量 DPPHラジカル捕捉活性を測定した 方 法 １ 試料 試料はアマゾンカムカム 株 より入手した10 および60 カムカム飲料を用いた 比較試料 として 緑茶 株 伊藤園 80 アセロラ飲料 株 ニチレイ を用いた 緑茶は茶葉１g に100mlの熱水を加え１分間抽出した通常飲用濃度を試料溶液とした ２ 試薬 1,1-diphenyl-2-picrylhydrazyl DPPH およびL-アスコルビン酸は和光純薬工業 株 より フェノール試液および没食子酸はナカライテスク 株 より購入した その他の試薬はすべて市 販の特級試薬を用いた ３ ビタミンＣ アスコルビン酸 の定量 ヒドラジン比色法 総アスコルビン酸の測定は試料溶液１mlに0.2 インドフェノール溶液0.1mlを添加して アス コルビン酸をデヒドロアスコルビン酸に酸化した後 ２ チオ尿素/ ５ メタリン酸１mlと２ 2,4-ジニトロフェニルヒドラジン溶液/9N硫酸0.5ml加え混和し 70 で30分間反応させた 反応 終了後 85 硫酸2.5mlを加えてよく混和し 室温で30分放置後 540nmにおける吸光度を測定 した なお 酸化型アスコルビン酸を測定する際は0.2 インドフェノール溶液の変わりに蒸留水を 添加し その他は総アスコルビン酸の測定と同様に操作した ４ ポリフェノールの定量 Folin-Denis法 ７ ポリフェノールの定量はFolin-Denis法に準じ 試験管に蒸留水2.7ml フェノール試薬0.2ml 試料0.1mlを加え混和後 10 Na2CO3溶液１mlを加えて全量を4.0mlとした よく混和後 室温 で30分間放置し 760nmにおける吸光度を測定した 別に没食子酸を標準物質として用い検量線 を作成し これより総ポリフェノール含量を算出した なお 0.1 ビタミンＣ溶液もポリフェ ノール含量 108±18mg/100ml として検出されるため 全ての試料のポリフェノール含量か らビタミンＣとして検出されるポリフェノール含量を差し引き 試料のポリフェノール含量を算 出した ５ DPPHラジカル捕捉活性の測定８ DPPHラジカル捕捉活性の測定は褐色試験管に100mＭトリス-塩酸緩衝液 ph ml 0.5mM DPPH/エタノール溶液２ml 試料溶液20-200µlを混和し 20分間反応 遮光下 させた 後 517nmにおける吸光度を測定した 80 アセロラ飲料および60 カムカム飲料は蒸留水で10 倍に希釈し 測定溶液とした システインを標準物質として用い １molのシステインが１mol
3 327 のDPPHを捕捉するとして DPPHラジカル捕捉活性 µmol DPPH trapped/100ml sample を 算出した ６ 全糖の定量 フェノール 硫酸法 糖の定量はフェノール 硫酸法で行った すなわち 試験管に試料溶液１ml ５ フェノー ル溶液１ml 濃硫酸５mlを加え混合後 室温になる前で放置し 490nmにおける吸光度を測定 した グルコース溶液を標準物質として検量線を作成し これより全糖量を算出した 結果および考察 １ ビタミンＣ含量 ビタミンＣ アスコルビン酸 は水溶性の抗酸化物質である カムカムは高いビタミンＣを含 んでいる そこで 試料中のビタミンＣ含量をヒドラジン比色法により測定し その結果を Fig. 1に示した ラベル表示と実測値はほぼ同じ値を示した ビタミンＣ含量は60 カムカム飲 料 80 ア セ ロ ラ 飲 料 10 カ ム カ ム 飲 料 緑 茶 の 順 に 高 か っ た 60 カ ム カ ム 飲 料 667mg/100ml は80 アセロラ飲料 400mg/100ml より高いビタミンＣ含量を示した Fig. 1 Vitamin C content of several drinks. Means with different letters are significantly different (p < 0.05). ３ DPPHラジカル捕捉活性 DPPHはエタノール溶液中で安定なラジカルを形成し ラジカルの消失とともにDPPHラジカ ルに由来する紫色が退色する性質を有している そのために近年DPPHを用いた比色法によるラ ジカル消去評価は 簡易な手法として汎用されている 本研究では 反応溶液の退色 すなわち 517nmにおける吸光度の減少が試料添加量に比例する範囲内において 試料溶液のDPPHラジカ ル捕捉活性を測定した その結果 カムカム飲料はDPPHラジカルを濃度依存的に消去した Fig. 3 いずれの試料も濃度依存的にDPPHラジカルを捕捉した 特に60 カムカム飲料と80 アセロラ飲料は強いラジカル捕捉活性を示し 原液では測定できないため 原液を10倍に希釈 して試料用液とした Fig. 4に各種飲料のDPPHラジカル捕捉活性を示した DPPHラジカル捕捉活性は60%カムカム 飲料 80 アセロラ飲料 緑茶 10 カムカム飲料 0.1 ビタミンＣ溶液の順に高かった 60 カムカム飲料 10,353µmol DPPH trapped /100ml sample は80 アセロラ飲料 5,694µmol DPPH trapped/100ml sample よ り 約 ２ 倍 高 い 活 性 を 示 し た 緑 茶 1,679µmol DPPH trapped/100ml sample 10 カムカム飲料 1,571µmol DPPH trapped/100ml sample および 0.1 ビタミンＣ溶液 1,189µmol DPPH trapped/100ml sample は同程度のDPPHラジカル捕捉 活性を示した カムカム飲料とアセロラ飲料の抗酸化活性にはビタミンＣおよびポリフェノール が 茶の抗酸化活性にはポリフェノールが関与していると考えられる

Means with different letters are significantly different (p < 0.05). ４ DPPHラジカル捕捉活性とビタミンＣ含量および総ポリフェノール含量との相関性 DPPHラジカル捕捉活性とビタミンＣおよび総ポリフェノール含量との相関性を得られたデー タを用いて解析した 横軸にビタミンＣ含量とポリフェノール含量 縦軸にDPPHラジカル捕捉 活性を表した Fig. 5 飲料のDPPHラジカル捕捉活性とビタミンＣおよび総ポリフェノール含 量の相関係数はr=0.9409で 飲料のDPPHラジカル捕捉活性とビタミンＣおよび総ポリフェノー ル含量との間に強い相関関係があることが示唆された

The data was plotted with DPPH radical-scavenging activity as ordinate and vitamin C and polyphenol content as abscissa. ５ 全糖量 フェノール 硫酸法による試料中の全糖量はFig. 6に示した 全糖量は80 アセロラ飲料 13g/100ml 60 カムカム飲料 10g/100ml 10 カムカム飲料 ７g/100ml の順に高か く 果汁濃度の高い順に糖濃度も高い値を示した これらの飲料は酸味を持っており 飲みやす くするために はちみつや果糖ブドウ糖液が添加されていると考えられる なお本実験の抗酸化 活性の測定法にこれらの糖は影響しないことも確認した Fig. 6 Glucose content of several drinks.
