Isolation and Characterization of Microorganisms Capable of Degrading (Ethylenediaminetetraacetato) Ferrate (III) Complex Hideo MIYAZAKI*, Seiji SUZUKI*, Atsuko TAKAHASHI* and Katsumi IMADA* Abstract The metal chelates of Ethylenediaminetetraacetic acid (edta) are not readily biodegradable. An attempt was made to isolate microorganisms which degraded chelating compounds such as edta from natural environment. Strain B-3 isolated from Fe (III) edta-containing soils was found to have the ability to degrade Fe (III) edta. A rate of degradation obserbed for NH4Fe (III) edta as the substrate was 52%, at 30 Ž with constant shaking for 3 days. Based on the numerical taxonomical studies, the strain was identified as Bacillus species. The three type cultures of Bacillus species also degraded Fe (III) edta. The degradation rates were dependent on species. OD: Degradation strain B-3 4 optical time course of Fe(III)edta by density 生 育 の 良 い6株 に つ い て 分 解 性 能 を 調 べ た 結 果 を 2に 示 す 6サ ン プ ル と も程 度 に 差 は あ っ た が,Fe(III) edtaに 対 し て 分 解 活 性 を 示 し た し か し,い も 1で 示 さ ず,ま 0.01%. た, み ら れ たedta 2Naを Fe(III)edtaに ずれの菌株 含 む培 地 で の増 殖 を 対 す る 分 解 率 も1に 掲 げ た値 よ り低 い もの とな った edta 2Naに 対 す る挙 動 が 土 壤 の 場 合 と分 離 した 菌 体 とで 異 な っ た 現 象 に つ い て は 次 の 理 由 を 考 え て い る す な わ ち 土 壤 サ ン プ ル の 場 合,edta 2Naを の 生 育 が み ら れ た の は,edta 2Naが と反 応 し てFe(III) edtaと 含 む培 地 で 菌 な り,edta 2Naの 微 生 物 に対 す る 毒 性11)が 無 く な る こ と に よ る も の で あ る 生 成 し た Fe(III)edtaは 漸 次 分 解 さ れ,見 か け 上edta 2 Naの 分 解 が 認 め られ た こ とに な る 序 で述 べ た 海 洋 性 微 生 物 の ス ク リ ーニ ン グ に お い て 顕 著 な分 解 活 性 を示 す細 菌 が 見 い 出 せ な か っ た の もedta 2Naを 含 む培 地 を用 い た こ と 1で はFe(III)edtaの う 値 で あ っ た の に 対 し,分 Electron micrograph of strain B-3 分 解 率 が64%と 3に 見 られ る ご と く,52%の 次 にFe(III)edtaに 分 解 率 を示 し た 馴 養 中 のB-3菌 を 用 いFe(III) edtaの 分 解 タ イ ム コ ー ス を37 の 静 置 培 養 に よ っ て調 べ た結 果 を4に 示す 時 間 を延 長 し て も分 解 率 が50%止 の は,分 解 が進 む に連 れpHが ま り とな っ て い る 上 昇 し て お り,こ の た め に 菌 の活 性 が 低 下 した もの と推 測 され る また,3で に起 因 し て い る 可 能性 が 考 え られ る ま た, Fig. 3 土壤 中 の鉄化合物 い 離 し た 菌 で は い ず れ も よ り低 は 同 条 件 下(静 置7日)の 4で は36%と 分 解 率 が25%で あ る の に対 し, 増 加 し て い る これ か ら は分 解 タ イ い 値 と な っ て い る こ れ に つ い て は土 壤 に含 まれ る栄 養 ム コー ス実 験 に用 い た 菌 体 の馴 化 が 進 んで き た可 能 性 が 素 の 効 果,或 考 え られ る も し馴 化 に よ りそ の分 解 能 も よ り高 くな る は 異 な る菌 が 共 存 す るが た め に 出 て くる協 こ とが 期 待 さ れ る な ら ば,Fe(III)edta濃 奏 効 果8)等 に よ る も の で あ る こ とが 考 え ら れ る 比 較 的Fe(III)edtaを て,0.01%Fe(III)edta濃 よ く 分 解 し た 菌 株B-3に つ い 度 で 分 解 度 を 調 べ た 結 果, 度 を次 第 に 高 め た 斜 面 培 地 に逐 次 移 植 す る こ とで よ り高 濃 度 のFe(III) edtaの 分 解 が 可 能 とな るの で,現 在 本 菌 株 の馴 養 を継 続
