2 2 反芻動物の GLP-1 分泌 8 図 1 にホルスタイン種子牛の出生後からの血漿 GLP-1 濃度の推移を示した 血漿 GLP-1 濃度は 出 生後から離乳に向けて徐々に減少したが 離乳を境 に 再び増加した 血漿グルコース濃度は出生後か ら徐々に減少し 離乳後 13 週齢では成牛と同程度 の濃度であった 一方で 血漿β ヒドロキシ酪酸 BHBA 濃度は 固形飼料摂取量の増加に伴い増 加し 離乳後は高値で推移した この BHBA 濃度 の増加は 反芻胃内での酪酸発酵に依存しているも のと推測される 離乳前の血漿 GLP-1 濃度は 単胃 動物と同様にグルコース濃度と類似した推移を示し たが 離乳後はグルコース濃度が低値で推移してい たにもかかわらず GLP-1 濃度が再び増加に転じた ことから 反芻動物ではグルコース吸収以外の要因 が GLP-1 分泌に関与している可能性が考えられ ま た酪酸発酵に起因する BHBA 濃度の増加と離乳後 の推移が類似していることから インスリン分泌同 様に反芻動物では VFA が GLP-1 分泌を促進する 一要因であると考えられた 図 2 および 3 に 離乳前後の子牛におけるグルコー スおよび VFA 頸静脈投与後の血漿 GLP-1 濃度の推 移を示した 単胃動物ではグルコース投与によって 血漿 GLP-1 濃度は増加するとされているが 離乳前 後の子牛では グルコースによる GLP-1 分泌促進は 認められなかった 図 2 本稿ではデータを示さな いが 離乳前の子牛に経口からラクトースを注入し た場合 血漿 GLP-1 濃度の増加は認めている Sugino et al., 16th AAAP proceeding, 2014 このことから 反芻動物においてもグルコース吸収が GLP-1 分泌促 進の一要因であると考えられるが 単胃動物のそれ と比較してその効果は弱い可能性が考えられる 一 方で VFA による血漿 GLP-1 濃度の増加は 出生 後から認められた 図 3 また 第一胃カニューレ 装着去勢ヒツジを用い 第一胃に VFA を注入した 場合も 同様に血漿 GLP-1 濃度の増加が示されてお り その効果は酪酸 プロピオン酸 酢酸の順で大 きかった 杉野ら 未発表 さらに 血漿 GLP-1 濃度は 消化吸収を反芻胃内発酵に依存している成 ヒツジにおいて 給飼後の増加が認められている 杉 野ら 未発表 このことから 反芻動物の GLP-1 36 図 1 子牛の血漿 GLP-1 a グルコース b およ び BHBA c 濃度推移 n 5 矢印 離乳 実線 給飼前濃度 点線 時系列 調整曲線
