P. mirabilisの 近 年,付 付 着 因 子 の解 析 着 に 関 与 す る菌 体 側 因 子 と して 線 毛 の 存 在 が 重 要 視 され て い る の で 臨 床 分 離P. mirabilisに お け る 線 毛 と各 種 細 胞 に 対 す る付 着 活 性 に解 析 を加 え た. Table 1に 示 した ご と く,臨 床 分 離 株 は 全 て ラ ッ トの 致 死 活 性,腎 形 成 を 示 し た.特 にNo. の 形 成 を認 め,そ 4-5mm径 測 定 に よ り求 膿瘍 お よび尿路 結石 6株 は 高 率 に 尿 路 結 石 の うちの膀胱 結石 の大 きさは P (Fig. 1 B)に 大 別 さ れ た. 動 物 通 過 後TSBに お け る 線 毛 の種 類,線 毛 密 度 は,ネ 作 為 に 各 々100個 選 び,電 子 顕 微 鏡 観 察 に よ り調 べ た.結 果 をTable 30は 他 の2株 P線 毛 保 有 菌 が23%, の臨床分 離株 に比べや や 高 い 致 死 活 性 を示 し た.ま た, No. 30に お い て は 腎 膿 瘍 が 高 率 に観 察 され た.一 に お い て は1匹 て い た が,致 だ け 約1mm径 方, No. 1 の 膀 胱 結 石 を有 し 死 活 性 お よ び 腎 膿 瘍 は 全 く観 察 さ マ ウ ス に お け るLD50の 測 定 か ら,臨 床 分 離 株 は 全 て108オ ー ター 以 下 の菌 数 を示 し,特 にNo. 2に 示 し た. No. 2は, 5 7 P線 毛 保 有 菌 が2%で, 各 線 毛 の 密 度 は 非 常 に低 か っ た. No. P線 毛 保 有 菌 の 割 合 が88%と 菌 は 存 在 しな か っ た.ま 6は, 5 高 く, 7 P線 毛 保 有 た, No. 6はNo. 2と 同 様 菌 の 個 体 当 りの 線 毛 密 度 は低 か っ た. No. 25 は, 98%が7 れ なか っ た. 目の各菌株 に ガ テ ィブ 染 色 を施 し た各 菌 株 を無 に達 す る もの が 多 く存 在 した. No. 25 お よ びNo. て 継 代3代 毛保 有菌 の割合 お よび線 P線 毛 を保 有 して お り,線 毛 保 有 菌 の 線 毛 密 度 も非 常 に 高 か った(Fig. 30は, 45%の 1 A). Fig. 2は,マ ウ ス 膀 胱 上 皮 細 胞 へ 最 も 良 く菌 が 付 着 した 例 を 示 した. 線 毛 の レ セ プ タ ー 認 識 能 の特 異 性 に 解 析 を加 え る ため,各 こ れ ら菌 株 に お い て 観 察 さ れ た赤 血 球 の 凝 集 性 は い ず れ も反 応 液 へ の マ ン ノー ス の 添 加 で 阻 害 菌 株 に お け る各 種 動 物 赤 血 球 凝 集 性 に検 討 を加 え た.血 球 凝 集 能 は4, 37 の 各 温 度 条 件 で 調 べ た. Analysis of pathogenic factors of Proteus mirabilis isolated from urinary tract infection Katsuhisa Department MUROTANI of Microbiology, Okayama University Medical School, Okayama 700, Japan (Director: Prof. Y. Kanemasa) Proteus mirabilis has several pathogenic factors such as adherent ability to urinary tract epitherial cells, urease, motility and resistance to urine. The pathogenic activities of clinically isolated P. mirabilis were analyzed. Higher pathogenic strains (No. 25 and No. 30) which had morphologically different pili but had a higher density of pili showed strong adherent activity to bladder epithelial cells of mouse and rat. These strains also showed a clear chemotaxis to urinary tract tissue extracts. These findings indicate that the combination of adherent, chemotaxis and urease activities is essential for causing of tipical kidney infection by P. mirabilis.
