We retrieved the mitochondrial DNA (mtdna) sequences and coding region information from 100 remains. Our results indicate that the Siberian input on the Ainu significantly affected their genetic structure, and that the Okhotsk people might be the main source of Siberian mtdna in the Ainu. These views were also confirmed by an analysis based on nonmetric cranial traits of the Hokkaido Ainu. 2 物学的な距離や系統関係は類推できるが 過去における疫病の流行や武力による衝突 集団間の移住や混血などといった 地域に限定された短期間の人口動態の変化を検出することは難しい したがって 限定された地域における集団の変遷史を明らかにするためには 現代人集団の解析のみならず 同地域の古代集団が持っていた遺伝的な特徴を明らかにする必要がある アイヌ集団に関してはこれまでも数多くの形態学的 遺伝学的な調査研究がなされてきた その中で アイヌは日本列島の基層集団の一つである縄文人との類縁性が認められ 彼らは縄文人の遺伝的影響を強く残した集団ではないかという仮説が提唱されている ただしこれまで行われた遺伝的な解析については 本土日本人との接触の影響を強く受けた現代人を対象としているため アイヌが本来持っていた遺伝的な特徴を捉えているとは言い難い 一方 近年の分子生物学の発展によって 古人骨由来の DNA の解析が可能になっており 江戸時代以前のアイヌ人骨を用いれば 彼らが過去に持っていた遺伝学的な特徴を明らかにすることも可能である 本研究は このような技術的な進歩をもとに これまで形態学的な研究のみによって追求されてきたアイヌ集団の人類学的な研究を更に深化させることを意図して計画された 2. 研究の目的北海道の先住民集団であるアイヌの形成史は 日本列島集団の形成を考える上でも重要で 近世アイヌ人骨についてはこれまでも多くの形態学的な調査研究がなされてきた しかし現在に至るまで 近世アイヌ人骨を対象とした集団遺伝学的な研究は存在せず, その遺伝学的な実体については知られていない そこで本研究では 明治以降の混血の影響を受けていない近世アイヌ人骨の形態および遺伝学的な研究を統合し 未だその実像が明確ではないアイヌ集団の成立史を明らかにすることを目的とした 3. 研究の方法札幌医科大学および北海道伊達市噴火湾文化研究所に所蔵されている近世アイヌ人骨の臼歯の歯髄から DNA を抽出し PCR 法を用いてミトコンドリア DNA の D-loop 領域および coding 領域を増幅し 配列決定を行った これによって得られた DNA データを用いて, アイヌの集団内および集団間の遺伝的な近縁性を検証した また頭骨の計測値と小変異の出現頻度データから同様の検討を行い 双方の知見を比較検討して その整合性を確認した 更に近隣の他集団との比較を通して アイヌ集団の成立史や集団としての特徴を 明らかにした DNA 抽出の方法は 現在のところサンプルからもっとも効率よく DNA を回収し 更に後の PCR 反応の阻害物質を排除できるとされているシリカマトリックスに DNA を吸着させる手法を用いた 実際には市販の DNA 回収キットのプロトコールに独自の改良を加えた方法を用いている 抽出した DNA をテンプレートとして ミトコンドリア DNA の D-loop 領域における塩基配列多型を解析した 我々のこれまでの研究により 北海道および東北の縄文人には N9b や D1a G1b など D-loop 領域の解析のみではハプログループを正確に決定できない系統が高頻度で観察されることがわかっている そこで ハプログループを特徴付けているミトコンドリア DNA の coding 領域の 1 塩基多型 (Single Nucleotide Polymorphisms; SNPs) についても併せて解析した D-loop 領域の塩基配列については 領域中特に多型性の高い hypervariable segment (HVS) Ⅰ および Ⅱ について 互いにオーバーラップする長さ 140~194 bp の PCR 産物が得られるプライマーを用いて DNA を PCR 法により増幅し 得られた各 PCR 産物の塩基配列をダイレクトシークエンス法により決定して HVS Ⅰ および Ⅱ の塩基配列を決定した DNA を採取した個体を中心に 頭蓋小変異 22 項目を調査し その頻度データをもとに これまで報告されている近隣集団との比較を行った 4. 研究成果札幌医科大学および伊達市噴火湾文化研究所が所蔵する近世アイヌ人骨 122 体を対象とした分析の結果 100 体から DNA 情報を得ることが出来た その結果 近世アイヌ集団から 13 種類のハプログループを検出したが 最も多かったのは オホーツク文化人に高頻度で見いだされているハプログループ Y だった (31%) それに続いて北海道の縄文人に高頻度で見られる N9b が検出され (19%) それ以外は北方系の先住集団に多い G1b,A4,Z などが検出された 一方 本土日本人に特徴的な D4 も 10% を越える頻度で存在し 更に南方の要素と考えられる B4,F1b,N9a なども頻度は少ないものの存在を確認できた これまでの研究で 北海道の縄文人にハプログループ Y が存在せず 主体をなすのは N9b であること これに対し オホーツク集団には高頻度で Y が存在することが知られている 従ってこの結果は 北海道のアイヌ集団は在来の縄文人の集団にオホーツク文化人が流入して構成されたというシナリオを支持している ただし 本土日本からの遺伝子流入も無視できないことも明らかとな

成果 本研究の解析で着目したのは 25 の遺伝性疾患とそれらの 57 の原因遺伝子で これらは ACMG が推奨する 偶発的 二次的所見としての遺伝情報の結果の返却を推奨する遺伝子のセットのうち常染色体上のものに相当し 大部分が遺伝性腫瘍や遺伝性循環器疾患の原因遺伝子です 本研究では 当機構で作成し
背景 全ゲノム解析や全エクソーム解析により ヒトゲノムの多様性の大量データが得られる時代となりました 当機構でも 日本人の全ゲノム解読を大規模に行い 検出された一塩基変異のリファレンスパネルを構築し 疾患と遺伝要因との関係解明のための研究に活用できるよう 国内外での利用促進を行っています 一方で ゲノム全体をカバーする解析は 疾患の原因遺伝子の塩基配列の解読もされてしまうこともあって 偶発的所見 二次的所見についての扱いのポリシーが課題になっています
